Hongos rizosféricos asociados a Citrus reticulata con capacidad antagónica hacia el patógeno del moho gris de la mandarina (Botrytis cinerea)

dc.description.abstractEl moho gris, causado por Botrytis cinerea, representa un problema fitosanitario relevante en el cultivo de mandarina debido a su comportamiento necrotrófico y a las pérdidas económicas que ocasiona. El uso continuo de fungicidas químicos ha favorecido la resistencia del patógeno y la acumulación de residuos, limitando su efectividad. Ante esta problemática, se tuvo como objetivo seleccionar cepas de hongos rizosféricos asociados a Citrus reticulata con capacidad antagónica frente a B. cinerea, como una estrategia alternativa de control biológico. Las muestras de suelo rizosférico fueron colectadas en cinco fundos dedicados a la producción de mandarina en la provincia de Cañete y posteriormente trasladadas al Laboratorio de Sanidad Vegetal de la Universidad Nacional de Cañete. El aislamiento de los hongos se efectuó empleando el medio Papa Dextrosa Agar (PDA), mientras que la identificación se realizó a nivel de género. Como resultado, se aislaron 56 cepas fúngicas, distribuidas en 18 géneros, entre los que se incluyen Acremonium sp., Aspergillus sp., Chaetomium sp., Cladosporium sp., Fusarium sp., Gliomastix sp., Humicola sp., Masoniella sp., Metarhizium sp., Microascus sp., Penicillium sp., Purpureocillium sp., Stachybotrys sp., Talaromyces sp., Thielaviopsis sp., Trichoderma sp., Paecilomyces sp., y Wardomyces sp. Del total de cepas aisladas, se seleccionaron 33 hongos rizosféricos para la evaluación de su capacidad antagonista frente B. cinerea. La actividad antagonista de las cepas de hongos rizosféricos se determinó mediante ensayos in vitro de antibiosis y micoparasitismo. La antibiosis se evaluó a partir del efecto inhibitorio de metabolitos secundarios sobre el crecimiento micelial de B. cinerea, mientras que el micoparasitismo se analizó mediante el método de enfrentamiento dual, cuantificándose el porcentaje de colonización del patógeno por los hongos rizosféricos El análisis estadístico de los datos se realizó utilizando los programas Infostat versión 2020 y Minitab versión 17. Los resultados obtenidos mostraron que los metabolitos producidos por las cepas HARZ-25 (Aspergillus niger) y HARZ-81 (Fusarium sp.) generaron los mayores porcentajes de inhibición micelial del patógeno con valores de 66,58 % y 63,29 % respectivamente. En contraste, la cepa HARZ-33 (Aspergillus pseudodeflectus) presentó el menor efecto inhibitorio con 3,85 %. Sin embargo, mediante el método del enfrentamiento dual, se evidenció que las cepas HARZ-54 (Trichoderma sp.) y HARZ-15 (Trichoderma asperellum) lograron inhibir el crecimiento del patógeno hasta un 73,97 % y 73,31 % respectivamente. Por el contrario, la cepa HARZ-53 (Humicola sp.) presentó el menor nivel de inhibición con 24,17 %. Asimismo, las siete cepas del género Trichoderma y una cepa de Fusarium lograron colonizar completamente al patógeno (100 %) demostrando un comportamiento como micoparásitos altamente agresivo. Los resultados obtenidos permiten concluir que los hongos rizosféricos nativos evaluados presentan un alto potencial como agentes de control biológico, evidenciando su capacidad para contribuir de manera efectiva y sostenible al manejo del moho gris de la mandarina. Su aplicación representa una alternativa técnicamente viable para disminuir la dependencia del uso de fungicidas químicos, favoreciendo prácticas agrícolas más seguras y compatibles con la conservación del ambiente.
dc.description.abstractGray mold, caused by Botrytis cinerea, represents a significant plant health problem in mandarin cultivation due to its necrotrophic nature and the economic losses it causes. The continued use of chemical fungicides has led to the development of resistance in the pathogen and the accumulation of residues, limiting the fungicides’ effectiveness. In light of this problem, the objective was to select strains of rhizosphere fungi associated with Citrus reticulata that exhibit antagonistic activity against B. cinerea, as an alternative biological control strategy. Rhizosphere soil samples were collected from five mandarin orchards in the province of Cañete and subsequently transported to the Plant Health Laboratory at the National University of Cañete. Fungal isolates were obtained using Potato Dextrose Agar (PDA), and identification was performed at the genus level. As a result, 56 fungal strains were isolated, distributed across 18 genera, including Acremonium sp., Aspergillus sp., Chaetomium sp., Cladosporium sp., Fusarium sp., Gliomastix sp., Humicola sp., Masoniella sp., Metarhizium sp., Microascus sp., Penicillium sp., Purpureocillium sp., Stachybotrys sp., Talaromyces sp., Thielaviopsis sp., Trichoderma sp., Paecilomyces sp., y Wardomyces sp. Of the total number of isolated strains, 33 rhizosphere fungi were selected to evaluate their antagonistic activity against B. cinerea. The antagonistic activity of the rhizosphere fungal strains was determined using in vitro antibiosis and mycoparasitism assays. Antibiosis was evaluated based on the inhibitory effect of secondary metabolites on the mycelial growth of B. cinerea, while mycoparasitism was analyzed using the dual challenge method, quantifying the percentage of pathogen colonization by the rhizosphere fungi. Statistical analysis of the data was performed using Infostat version 2020 and Minitab version 17. The results showed that the metabolites produced by strains HARZ-25 (Aspergillus niger) and HARZ-81 (Fusarium sp.) produced the highest percentages of mycelial inhibition of the pathogen, with values of 66.58% and 63.29%, respectively. In contrast, strain HARZ-33 (Aspergillus pseudodeflectus) exhibited the lowest inhibitory effect at 3.85%. Furthermore, the seven strains of the genus Trichoderma and one strain of Fusarium were able to completely colonize the pathogen (100%), demonstrating highly aggressive mycoparasitic behavior. The results obtained lead to the conclusion that the native rhizosphere fungi evaluated show high potential as biological control agents, demonstrating their ability to contribute effectively and sustainably to the management of gray mold in mandarins. Their application represents a technically viable alternative for reducing dependence on chemical fungicides, promoting agricultural practices that are safer and more compatible with environmental conservation.
